Intraspecies differences in cold hardiness, carbohydrate content and β-amylase gene expression of Vaccinium corymbosum during cold acclimation and deacclimation.

چکیده
Changes in cold hardiness, carbohydrate content and β-amylase gene expression were monitored in the shoots of the highbush blueberry (Vaccinium corymbosum L.) cultivars 'Sharpblue' and 'Jersey' during cold acclimation (CA) and deacclimation (DA). The seasonal patterns were similar in both cultivars, but the levels of cold hardiness determined by electrolyte leakage analysis were significantly different; 'Jersey' was hardier than 'Sharpblue'. Cold hardiness was closely related to total soluble sugar content (r = -0.98** and -0.99** for 'Sharpblue' and 'Jersey', respectively). In 'Jersey', more soluble sugars accumulated during CA. Of the detected soluble sugars, glucose, fructose and raffinose contents were significantly associated with cold hardiness in both cultivars. Sucrose was abundant in both cultivars, and stachyose content changed significantly during CA and DA. However, they were not associated with cold hardiness. A sharp decrease in starch contents in the middle of CA coincided with β-amylase gene (VcBMY) expression, indicating the conversion of starch into soluble sugars. During CA, VcBMY was expressed up to twofold higher in 'Jersey' than in 'Sharpblue'. These results suggest that intraspecies differences in the cold hardiness of highbush blueberries are associated with total soluble sugar content, which is driven partly by differential expression of VcBMY.

Deacclimation and reacclimation of cold-hardy plants: Current understanding and emerging concepts
The abilities of cold-hardy plants to resist deacclimation during transient warm spells and to reacclimate when cold temperatures return are significant for winter survival. Yet compared to the volume of research on the biology of cold acclimation, relatively little is known about how plants maintain and/or reacquire cold hardiness in late winter and spring. This review summarizes the past 40 y...
متن کاملMajor diVerences observed in transcript proWles of blueberry during cold acclimation under Weld and cold room conditions
Our laboratory has been working toward increasing our understanding of the genetic control of cold hardiness in blueberry (Vaccinium section Cyanococcus) to ultimately use this information to develop more cold hardy cultivars for the industry. Here, we report using cDNA microarrays to monitor changes in gene expression at multiple times during cold acclimation under Weld and cold room condition...
متن کاملBud Hardiness and Deacclimation in Blueberry Cultivars With Varying Species Ancestry: Flowering Time May Not Be a Good Indicator of Deacclimation
Blueberry cultivars with varying percentages of species ancestry (V. corymbosum L., V. angustifolium Ait., V. ashei Reade, V. darrowi Camp) were assayed in mid-February to determine initial bud hardiness, and rates of deacclimation under constant temperature conditions. The LT50 (the temperature at which 50% lethality occurs) of detached shoots of field-grown plants of ‘Weymouth’, ‘Bluecrop’, ‘...
متن کاملTime-dependent deacclimation after cold acclimation in Arabidopsis thaliana accessions
During low temperature exposure, Arabidopsis thaliana and many other plants from temperate climates increase in freezing tolerance in a process termed cold acclimation. However, the correct timing and rate of deacclimation, resulting in loss of freezing tolerance and initiation of growth is equally important for plant fitness and survival. While the molecular basis of cold acclimation has been ...

Freezing injury is an important limiting factor in the production of pomegranate in Iran. The aim of the present study was to evaluate the effect of paclobutrazol (PBZ) on cold hardiness of pomegranate (Punica granatum L.) cv. ‘Malas Saveh’. Different concentrations of PBZ including 0 (control), 31, 62, 125 or 250 mg L–1 were sprayed on one-year-old plants in August and the electrolyte leakage o...
